Four games are set to be play on the league's opening day, with Shelbourne and Cork City getting things underway in Tolka Park at 2pm.
The season was originally due to kick off in March but was delayed due to the pandemic.
The season will now go ahead with nine clubs vying for the top spot, with Peamount United hoping to retain their title.
The league will be broken into two 'phases'. In phase one, each team will play the other clubs once, before being split into two groups for phase two, in which teams will play each other again.
Saturday evening, Galway will play DLR waves at 6pm with the two remaining games, Peamount against Treaty United, and Wexford Youth against Bohemians kicking off at 6.30pm.
Athlone Town will lie in wait, having a bye in the first round of action.
